RESEARCH SUMMARY
This work introduces self-immolative polyglyoxylamide (PGA) hydrogels capable of traceless drug release via cleavage-triggered polymer backbone degradation. Through tunable crosslinking and pendant linker chemistry, the hydrogels allow covalent drug immobilization followed by triggered release under physiological stimuli. Mechanical and swelling tests demonstrated excellent structural integrity and reversible degradation behavior. The platform offers potential for smart drug delivery systems where payload release is mediated by chemical or enzymatic signals without residual linker fragments.

CELLSCALE INSTRUMENT USED

UniVert

Compressive mechanical testing of cylindrical PGA hydrogel samples was conducted using a CellScale UniVert mechanical testing system (Waterloo, ON, Canada) equipped with a 10 N load cell. Samples were equilibrated in PBS at 37 °C and compressed to 40 % strain under unconfined conditions. Compressive moduli were calculated from the 5–15 % linear strain region, demonstrating tunable stiffness between 20–80 kPa depending on crosslink density and drug conjugation state.
