RESEARCH SUMMARY
This preclinical study evaluated the in vivo functionality and remodeling behavior of bioresorbable, pliable elastomeric heart valve prostheses fabricated from bis-urea-modified polycarbonate (PC-BU). Implanted in the aortic position of sheep for up to 12 months, the valves were assessed for durability, calcification, cell colonization, and tissue formation. Most valves remained pliable and structurally intact, free of thrombotic or calcific complications, though some exhibited cusp thickening or perforations. Histological analysis revealed limited cell infiltration and collagen deposition mainly in hinge regions, with minimal scaffold resorption. These findings demonstrate partial success in creating functional in situ tissue-engineered aortic valves, highlighting the challenge of achieving balanced tissue regeneration under systemic pressure.

CELLSCALE INSTRUMENT USED

BioTester

The mechanical behavior of explanted valve leaflets was evaluated using a CellScale BioTester 5000. Square samples (5 × 5 mm²) from the tip and belly regions of the valves were subjected to biaxial tensile testing in radial and circumferential directions using a 1.5 N load cell and LabJoy software v8.01. Strain was incremented in 5% steps up to 40%, with nine preconditioning cycles and one measurement cycle per step. Stress–strain curves and tangent moduli at 35% strain showed similar behavior to pre-implant materials, confirming mechanical stability after 12 months in vivo.
