Effect of Skeletal Muscle Immobilization on Regional Anisotropic Viscohyperelastic Properties Change in a Rat Model

RESEARCH SUMMARY
This study investigated the regional effects of skeletal muscle immobilization on anisotropic viscohyperelastic behavior using an in vitro rat model. Equibiaxial relaxation tests were performed on distal, medial, and proximal regions of immobilized biceps brachii samples to quantify regional differences in stiffness and relaxation. The results showed that immobilization primarily affected viscoelastic parametersโ€”leading to increased relaxation coefficientsโ€”while the excising zone determined hyperelastic stiffness. Histological analysis revealed significant atrophy and fibrosis characterized by increased perimysium and endomysium collagen fractions. These mechanical and structural correlations suggest that immobilization-induced remodeling alters muscle anisotropy and viscoelasticity, with implications for spastic myopathy and rehabilitation research.

CELLSCALE INSTRUMENT USED

BioTester

Equibiaxial relaxation tests were conducted using a CellScale BioTester with orthogonal traction axes. Biceps brachii samples were mounted in a saline bath at 26 ยฑ 1 ยฐC and subjected to 30 % biaxial strain followed by a 300 s relaxation phase. The BioTester precisely controlled preloading (30 mN per axis) and tracked stressโ€“strain responses, enabling identification of viscohyperelastic parameters via anisotropic strain energy and Maxwell modeling. This setup provided quantitative insights into how short-position immobilization altered muscle stiffness and relaxation dynamics.
