Acetylation of alginate enables the production of inks that mimic the chemical properties of P. aeruginosa biofilm

RESEARCH SUMMARY
This study investigates how acetylation alters the physicochemical and mechanical properties of alginate hydrogels, creating tunable biofilm-like materials that mimic *Pseudomonas aeruginosa* extracellular matrices in cystic fibrosis infections. Acetylated alginate with varying degrees of substitution (10–36%) showed altered viscosity, reduced calcium ion affinity, and pH-dependent softening, aligning with mucoid alginate secreted by *P. aeruginosa*. The acetylation broadened the linear viscoelastic region, reduced Young’s modulus, and modulated antibiotic permeability, offering an engineered hydrogel platform that reproduces bacterial biofilm mechanics and drug transport behavior.

CELLSCALE INSTRUMENT USED

MicroTester

Compression tests were conducted using a CellScale MicroTester in compression mode on Ca²⁺-crosslinked alginate microbeads at pH 6.5 and 7.0. The force–displacement data were analyzed with the Hertz contact model to determine Young’s modulus, which ranged from 9.8–12 kPa at pH 7 and decreased to 5–7.5 kPa for highly acetylated samples. The MicroTester confirmed that increasing acetylation reduces mechanical stiffness by lowering calcium crosslinking density, providing quantitative evidence of biofilm-like compliance.
