A ‘Spicy’ Mechanotransduction Switch: Capsaicin-Activated TRPV1 Receptor Modulates Osteosarcoma Cell Behavior and Drug Sensitivity

RESEARCH SUMMARY
This study investigates how TRPV1, a mechanosensitive ion channel activated by capsaicin, regulates osteosarcoma cell behavior and chemosensitivity. Using the MechanoCulture FX to apply uniaxial cyclic stretch (0.5% elongation at 1 Hz for 24 h), researchers demonstrated that mechanical strain and chemical TRPV1 activation both induced subtype-specific responses in osteosarcoma cell lines (U-2 OS and SAOS-2). Mechanical loading or capsaicin treatment triggered reorientation, cytoskeletal remodeling, and changes in adhesion and migration through the Src–acH3 mechanosignaling axis. TRPV1 antagonism with AMG9810 abolished these effects, confirming TRPV1’s key role in mechanochemical signaling. The work positions TRPV1 as a mechanosensitive hub integrating mechanical and biochemical cues to control osteosarcoma aggressiveness and drug response.

MechanoCulture FX

The MechanoCulture FX bioreactor was used to apply cyclic uniaxial stretch (0.5% elongation, 1 Hz) for 24 hours to U-2 OS, SAOS-2, and hFOB cells. Cells were cultured on deformable silicone plates, mimicking physiological strain conditions. This controlled mechanical loading enabled researchers to evaluate TRPV1-dependent mechanotransduction pathways, cytoskeletal reorganization, and drug response under reproducible mechanical stress.
