Ketoprofen-Based Polymer–Drug Nanoparticles Provide Anti-Inflammatory Properties to HA/Collagen Hydrogels

RESEARCH SUMMARY
This study developed ketoprofen-based polymer–drug nanoparticles incorporated into hyaluronan/collagen hydrogels and cryogels for use as anti-inflammatory wound dressings. The composite biomaterials were designed to modulate macrophage-mediated inflammatory responses while supporting tissue regeneration. Mechanical characterization demonstrated that hydrogel and cryogel stiffness could be tuned through formulation and processing, influencing macrophage adhesion, activation, and cytokine production. In vitro studies showed that nanoparticle-loaded matrices reduced nitric oxide release and multinucleated giant cell formation while maintaining cell viability, highlighting the importance of scaffold mechanics in regulating immune response and wound healing outcomes.

CELLSCALE INSTRUMENT USED

MicroSquisher

Unconfined compression testing was performed using a CellScale MicroSquisher to quantify the elastic modulus of hyaluronan/collagen hydrogels and cryogels used as wound dressing matrices. Cylindrical specimens (3 mm diameter) were compressed under displacement control to generate stress–strain curves in the low-force regime, enabling accurate measurement of soft material stiffness. These mechanical measurements were used to correlate scaffold mechanical properties with macrophage behavior and inflammatory response, providing insight into how matrix mechanics influence wound healing and fibrosis-related processes.
