From Bench to Chairside: Collagen Scaffolds in Combination with Mesenchymal Stromal Cells for Gingival Augmentation

RESEARCH SUMMARY
This study compared three collagen-based xenogeneic scaffolds—Fibro-Gide, FibroMATRIX, and Mucoderm—for gingival augmentation, both alone and combined with mesenchymal stromal cells (MSCs). Mechanical, structural, and in vivo analyses revealed that FibroMATRIX provided an optimal balance between stiffness, porosity, and bioactivity. Micro-CT, indentation, and histology demonstrated that FibroMATRIX supported greater vascularization and tissue integration than Fibro-Gide or Mucoderm. FibroMATRIX + MSCs yielded higher lamina propria thickness and vessel density, while Fibro-Gide inhibited angiogenesis due to dense architecture. Results demonstrate that scaffold stiffness and porosity critically influence gingival regeneration outcomes.

CELLSCALE INSTRUMENT USED

MicroTester

Mechanical indentation was performed using a CellScale MicroTester G2 (Waterloo, Canada) in displacement-controlled mode. Hydrated collagen scaffolds were tested in PBS using a 12 N/m cantilever and 500 µm spherical tip to depths of 300–400 µm at 30 µm/s with a 30 s hold period. Indentation data were analyzed using Hertz and Lee–Radok models to calculate instantaneous (E₀) and equilibrium (E∞) moduli. FibroMATRIX and Fibro-Gide exhibited similar stiffness, while Mucoderm was ~4× stiffer. The MicroTester G2 allowed precise quantification of scaffold stiffness and viscoelastic properties relevant to tissue integration.
